Effect of NO−Precursor (L−Arginine) and NO−Synthase Inhibitor (L−NAME) on the Secretion of Thyroid Hormone and Thyrotropin (TSH) in Female Rats

چکیده
Background. Nitric oxide (NO) plays an important role in various biological processes. It has been suggested that NO may modulate the functions of the endocrine system. The aim of this study was to examine the role of nitric oxide in total triiodothyronine (T3), free thyroxine (fT4) and TSH secretion in adult female Wistar rats. Material and Methods. The animals were treated with L−NAME (10 mg/kg IP) and L−arginine (100 mg/kg IP) daily for 7 consecutive days. On the day 8th plasma TSH concentration was estimated by immunoenzymatic micro− particle method, while thyroid hormones, T3 and fT4, were determined using radioimmunoassay method. Results. The plasma TSH concentration was very significantly diminished, following L−arginine treatment. On the other hand L−arginine was without effect on T3 and fT4 plasma level. L−NAME administrated for 7 days did not produce any significant changes in the plasma concentration of T3, fT4 and TSH. Conclusions. These findings may indicate that long−term L−arginine exposure decreases TSH secretion under ba− sal condition in female rats. However, the results of this study need further investigation that might be able to con− firm whether L−arginine action on TSH secretion is indeed mediated via NO (Adv. Clin. Exp. Med. 2004, 13, 1, 23–26).

The effect of L-arginine and L-NAME on intravenous self-administration of morphine in rats
The involvement of nitric oxide (NO) in the reinforcing properties of opiate reward was studied by examining the effect of a NO precursor (L-arginine) and a NO synthase inhibitor (L-NAME) on the rate of intravenous self-administration of morphine. This experiment was investigated in male albino Sprague Dawley rats (300 ± 50 g). At doses of 5 and 10 mg/kg (i.p.), L-arginine decreased morphine ...

Introduction: The current study aimed at investigating the role of Nitric Oxide (NO) in the maintenance of anxiety and depression induced by stress in male Wistar rats using intra-Basolateral Amygdala (BLA) injection of NO precursor, L-arginine, Nitric Oxide Synthase (NOS) inhibitor, and L-NAME. Methods: Two 23-gauge stainless steel cannulas were placed in the BLA, stereotaxically. Seven days...

Introduction: This study was performed to evaluate the effects of intra-nucleus accumbens shell microinjection of Larginine (NO precursor) and L-NAME (nitric oxide synthase (NOS) inhibitor) on morphine withdrawal signs in male rats. Method: Rats were anaesthetized with a combination of ketamine and xylazine, and placed in stereotaxic apparatus, and a guide cannula was inserted into nucleus accu...
متن کاملThe effect of intraaccumbal injection of L-arginine and L-NAME on induction and removal of conditioned place preference in rat
Nucleus accumbens (Nac) has been considered as a center for the induction of drug dependence. Since a high concentration of the enzyme, nitric oxide synthase (NOS) has been found in the Nac, and this fact that the role of nitric oxide (NO) in acquisition and expression of drug dependence has become clear in recent years, therefore in this study the effects of intraaccumbal injections of L-argin...
متن کاملIncrease of lead-induced release of N-acetyl-p-D-glucosaminidase by NO synthase in perfused kidney of rat
Urinary N-acetyl-β-D-glucosalninidase (NAG) has been proved to be a useful marker of early renal injury as a result of factors such as lead toxicity. In this study the effect of lead acetate on the kidney and its correlation with the nitric oxide (NO) system was investigated by determining the NAG release in perfused kidney of rat. Lead acetate caused a time- and dose-dependent increase in en...
